Just bought a 2006 EX about a month ago, and this weekend I tried to use the key for the first time to open the door, and it wouldn't go in! Never thought to try the key in the door before purchase!

I've got the 2005-2006 service manual so I can see how to take the panels off and remove the cylinder. I'm guessing there are two possibilities:
1) Something is jammed in there, like a pick (!) or something that broke off
2) The tumblers/plates/cylinder are all messed up

First thing I should do is remove the cylinder to examine it and see what the problem is. If it's not the simple case of something jammed then will I need to order a new cylinder? I believe it would be part #72185-SHJ-A01. I can either take the cylinder and key to a dealer or locksmith to get it rekeyed or maybe do it myself if the plates/tumblers aren't damaged?

Thanks for any info!

I would start with the easy stuff first. Get a good lock lubricant and spray it in the cylinder to flush out 5 years of gunk. It's likely the lock has never been used and could be full of dirt/sand/salt/etc.
